Only with manual ads. With Auto-Ads you can not chose where Adsense places them, but you can exclude areas from the Adsens dashboard (Adsense website)
I don’t think there is anything Hugo specific here. It’s Adsense related.

You will need to create left and right sidebars with the layout to place your manual ads as partials or pick a theme that can support what you want.
